As of April 15, I had only been admitted to one MD/PhD Program and I let that program know that I was "planning to attend" (since this program starts rotations in June). However, yesterday I was taken off of a "hold" status at another school and admitted. Does this mean that I have two weeks from yesterday to decide which school I want to go to? Also, do I need to directly contact the first school that I received another admissions offer as of yesterday, so I am now reconsidering?

If you received a valid offer, your original MD/PhD program will realize that you still have multiple acceptances in their acceptance report. You will risk rescindance from the acceptance from that original program, you should contact your current acceptance school and tell them what happened. If they are fair, they will let you have several days but "less than two weeks." I suspect than a week is what is fair given that they are starting in June. However, you should contact them soon...

The program that offer you must register that offer in AMCAS within 7 days (see #2 of TR for PDs).

Addendum:
Keep in mind that from the standpoint of the program that accepted you first, you should be down to a single choice by April 15. New options available to you after that deadline might result in them scrambling to fill that slot, often with lesser success than withdrawals during the first 2 weeks of April. The real problem for MD/PhD programs starting in June are the withdrawals on May 15, they can lose an accepted student with a late acceptance from somebody else. Often, that will result in a slot not filled, or filled later with somebody from the MS-1 or MS-2 class.
