Welcome to Part 6! Everyone did a bang up job by hitting 10k in less than 12 days--11 days 19 hours and 18 minutes to be exact. A new record! 😀

For those not familiar with what we do here, Molly has been kind enough to put together a nice description.

One of the other hostesses says "Bitch, please," a lot, but she's joking... don't take it personally.

Forget anything you've heard about the girl who died here from arsenic poisoning. That was a long time ago, and she's still alive, anyway... just busy so she doesn't come in as much. Besides, a little arsenic adds a lot of flavor to just about any dish.

Pretty flowers by the entrance, hey? Lilis, they are. Beautiful. And the music is totally hopping... we have our very own DJ. Looks a little like Brad Pitt.

I guess that's pretty much all you need to know. I'm usually in the corner booth over there drinking a nice glass of wine. You're welcome to join me, anytime. We're open 24/7.

I think I need another cup of coffee....didn't get a goodnights sleep.....my dog woke me up last night 'cause she had an upset stomach.....it was making a screeching sound.....sounded like an alien in there, lol.

I've been trying to get football tickets 'cause my husband has been wanting to see a football game live but it's sold out for the whole season, wtf!! 🙁 I have to get on a waiting list.

Going to snarf down some lunch and work on my talk for tomorrow while I have a page-free moment here. And have to prepare for surg path conference tomorrow.

Hi randomite peeps! Can I get some mad biochemistry knowledge from you guys? I am giving this chemistry talk tomorrow and am talking about the Km of this enzyme. (I had to look this up. 😳 Now I vaguely remember that it is the concentration of substrate leading to half-maximal velocity).

So: question: how so you "say" this in a talk? Is it just K M? or K sub M? or is there some word that I should say? I don't want to sound like a *****. I don't think I have said or thought about Km for about 7-8 years.

Its generally represented Km with an uppercase K and a lowercase m

The Km is the substrate concentration at which you reach 1/2 the maximum velocity and is generally that point where the linearly increasing part of the michelis menton curve starts to end and create a plateau.

The lower the Km the higher the substrate affinity.

Km will not be affected if there is a noncompetitive inhibitor but the Vmax will decrease. If the inhibitor is noncompetitive, then the inhibitor binds at a place other then the active site so more substrate won't over turn its effects. Hence, Km will not be altered.

On the other hand, if you have COMPETITIVE inhibition then you can override the inhibitor by adding more substrate. Hence, competitive inhibition will increase the Km value but the Vmax will remain the same because a competitive inhibitor mimics the transition state of the original substrate.

Oh and what I just described is michelis menton kinetics also known as saturation kinetics. It works for single subunit enzymes.

If enzymes are multisubunit then they operate via sigmoidal kinetics (cooperativity).

I think I've always heard it pronounced as Km not Ksubm. That's probably just some biochem speak.

I guess it is represented as K sub M in the book but i've also seen it represented as Km is other sources.

I guess I'd use KsubM in the written part but pronounce it Km.

I don't know what's gotten into me recently, but I've changed the music I've been listening to. i.e. the 1999 Big Tymers stuff, lol. MJ's Thriller and even Seal's Kiss from a Rose...LMAO!

Why would the school I am applying to want official transcripts now? 😕 Do they usually ask for that even before an interview?

And I officially HATE organic chemistry lab. It's the first class I have truely hated since my psychology of prejudice class way back in 1999. UGH!!! They give us way too much to do in 3 hours and our experiment today didn't even work right because the TA didn't even know how to do it and gave us wrong instruction. 😡

hey! i'm doing ok...also busy at work, but I think that with the anticipated 800 subjects coming in for our study, I SHOULD be busy! 😛

i dunno about schools asking for transcripts before interviews, but Miami asked for my official transcript at the same time they asked for a secondary. So it's not unheard of.

You have the HEAT on?!?! Are you crazy? I never turned the heat on ONCE at my apt in north Florida. :laugh:

If you think it was warm, it wasn't. I'd leave my window open overnight when it was 40 degrees (forty!!!). It was awesome. It got down to 50 degrees once on the thermostat in the center hallway (warmest part of house). 😀 I loved every bit of it.

Oxeye, sorry to hear you don't like orgo lab. 🙁

my roomate wanted the heat on. i have a fleece blanket and down comforter so i don't need the heater on. my parents keep their house at 65 in the winter so i'm used to it. but it did have to get fixed. the pilot light was off.

my roomates at school were insane. one girl insisted that it was 75 in the winter and 60 in the summer. i was sick year round because of her

crap i meant 85. i had to reason with her to get it into the 70s though. insane. INSANE. she had a broken vent, so that heat was constantly on.... ugh ugh ugh
