I applied to a bunch of schools, got rejected right off the bat at Vanderbilt, then didn't return secondaries to most schools I applied to except for WFU, UNC, ECU. My reasoning was that if I got into a school i really didnt want to go to (I really only wanted to go to UNC) then I was just going to wait another year, travel more, and apply again. Wake forest put me on some kind of quasi interview waitlist where I had to write some kind of letter in order to get an interview, which I didnt do. I got straightup rejected from ECU, but I basically told them I didnt want to do rural medicine.
So long story short, UNC was the only waitlist I was on and I didnt have any other offers pending.
My 'applicant profile' is:
School: Duke
MCAT: VR: 9 PS: 12 BS: 12 (33 total)
GPA: 3.3
Sci GPA 3.2
Extras: I'm from Chapel Hill, volunteered then worked paid as an EMT (up to 95hrs/week) during my Junior/senior years and during my year off, had about 1.5 years lab experience, led a bunch of 2week backpacking/climbing trips for freshman, taught a house course on wilderness survival/philosophy stuff, 1 year as news/sports photographer, and taught photography to kids in hawaii for 5 weeks (hehe)
My interview went decently well w/ both interviewers.
I wrote a one sentence letter (sent snail mail) before I got on the waiting list saying that I would definitely go to UNC if I got in and that it was my number 1 choice. I called twice to the Admissions Office about stupid questions after I got in off the waiting list, nothing that should have helped me get in.
I posted info below from our health prof advising center's website that could help you in case you didnt already hear this advice somewhere else. This was written by our Dean of Health Professions Advising, very well respected lady and knows most of the Deans of Admissions, so she knows her ****.

WAIT LISTS
If you are on a wait list, there are some proactive steps you can take that might improve your chances of being admitted to that school.
You should write a letter to the Dean/Director of Admission and express your interest in the school. Say something specific about why you are interested in the school. Perhaps they have a particular program or aspect of their curriculum that you think is exciting or a good match for what you are looking for in a school. Then keep in touch with the school every 3 or so weeks. If you do this in writing, you can feel more confident that it will be noted in your file. However, some schools keep track of phone or email communication.
If you have had any additional experience, honor, award, etc., then notify the school in writing. Be sure to send them an updated transcript if you have taken additional courses.
If there is someone who could write a letter of support for you now who did not write previously, or if there is someone who wrote for you before but now knows you better, that person should write a letter specifically the school or schools where you are on a wait list.
Finally, if you feel that a school is your first choice (and you would withdraw from another school if you are holding an acceptance), the tell the school that in writing. But remember, you can only say that to one school. And be sure not to make promises you can't keep.
