WHAT? I feel like my world has been turned upside down. There are very few things I can count on in life, and having the mushinator as the top poster was one of them.

I think we are just getting the new SDN froshes for next cycle trickling in now and our world as we knew it will soon be lost forever. All good things must come to an end I suppose, we'll just have to adapt 😉

Fortunately images are for the most part screened out on my work computer, so I was spared the visual until I got home :laugh:
 
Severus, to address your concerns about post deletions. My understanding is, after some duration previous versions of large multipart threads are being deleted at some point after it is closed and restarted.

So I would expect if you post in those types of thread often, eventually your post count will be effected.

Is this a major concern for posters of these types of threads? If it is, I'll certainly mention it to someone with more power than me, and see if there is any solution, either by just keeping your post count, or by somehow archiving old threads.

I'm shocked the NIH's filters don't screen out the naked men. People at work probably think I am a nympho. Thanks guys. Appreciate it. 😛

I still cant figure out how to post pictures without them coming out as thumbnails.
 
For me DKMs pic showed up in the middle of the medical library with all the med students around me looking. The pink font made me that much better too.

After weeks of failure, I just optimized a reaction to use half of the reagents that we were using before.. The girl before me tried it and never got it to work. Seeing as how I run hundreds of these reactions a week, we are going to save a ton of money. In short, I am awesome. See, I really do accomplish things at work.. I only waste time on SDN between experiments =)

Now all I have to do is set up another 6 plates of PCR reactions before I go home.. 😴

Way to go! PCR optimization can be a real pain. I empathize with you. Those reagent kits are seriously expensive. When doing direct sequencing off my PCR products, I used to dilute the big Dye kits by half, but it got sketchy going much more then that at times.

I started work in September... At one point in December I counted from my lab notebook all the times I ran PCRs and it was like 80 something.

I have to run duplex PCRs and that was so hard to optimize cuz my internal control gene loved to fluctuate all over the place giving me weak bands with one target gene and then uber strong ones with others. And i had like 25 targets to go thru!!
 
I singlehandedly make up my lab's sequencing core. Which means I have the most boring job ever. You know this is true because there are ROBOTS THAT DO MY JOB. Its just cheaper to pay me. So instead of getting a research project, my PI is giving me lab technician work (I am not "too good" to be a lab tech, but they get paid way more than post-bacs..). I have no chance of publishing and I hate sequencing. BTW, this is not what I was told I would be doing when I agreed to take the job. But I can't find another job now, just in case I do move away for school (if I get in - still hoping- kinda). So I am screwed for the time being. Its not too bad I guess - I could still be waiting tables.. At least the people I work with are pretty cool. and I get to work at the NIH. 🙂
 
for those of you in the DC area -

Scrubs is on channel 20 at 6 and 6:30 every week night (i think), and for those with cable, its also on Comedy Central at 7 and 7:30. Additionally, the Sopranos is on at 9 on A&E on Wednesday and repeated the next Monday at 9. I can't believe I missed the Sopranos craze - its such an awesome show.. I know what I want for my birthday... 😀

Is it bad that I park myself in the front the tube to take my mind off of med school stress?
